Novel RA drug shows promise in late trials

Tasocitinib, a novel oral Janus kinase inhibitor for rheumatoid arthritis, has shown promise in late stage clinical trials. Data has been released by manufacturer Pfizer from Oral Solo, a six-month phase III double blind randomised controlled trial of 611 patients with moderate to severe RA who had an inadequate response to at least one disease-modifying antirheumatic drug and received tasocitinib 5mg, 10mg or placebo, twice daily.

After three months, the ACR20 response rates (20 per cent improvement on the American College of Rheumatology scale) were 59.8 per cent for the 5mg dose, 65.7 per cent for the 10mg dose and 26.7 per cent for placebo. There was a similar frequency of adverse events across the groups, with serious adverse events occurring in 4.1 per cent of patients. Further results from an ongoing phase II/III trial, Oral Sequel, show that the most frequent adverse events are infections.
